Abstract: The invention belongs to the technical field of plant gene engineering and relates to a ribonucleic acid (RNA) interference vector of a cotton fiber specific arabinogalactan protein glycosyl transferase GhGalT1 gene, and a construction method and application thereof. The RNA interference vector comprises a skeleton vector and a hairpin structure, wherein the skeleton vector contains a cotton fiber GhRDL1 promoter core sequence; the hairpin structure contains a GhTUA6 gene intron, a positive-sense strand and an anti-sense strand; and the positive-sense strand and the anti-sense strand are tandem sense strand nucleotide sequence and anti-sense strand nucleotide sequence of the specific deoxyribonucleic acid (DNA) fragment of the cotton GhGalT1 gene. The GhGalT1 RNAi interference vector constructed by the construction method provided by the invention can be applied to cotton fiber quality improved molecular breeding, so the expression level of the GhGalT1 gene of the GhGalT1 RNAi transgenic cotton fiber is obviously inhibited to different degrees, the length of mature fiber is significantly increased, the fiber quality is obviously improved, and stable inheritance is realized.
